TDP-43 mislocalization and aggregation occur in more than 97% of ALS cases, about 40% of frontotemporal dementia and a substantial proportion of Alzheimer's-spectrum disease, either as co-pathology or primary pathology (Limbic-predominant Age-related TDP-43 Encephalopathy,LATE). Despite its central role across neurodegenerative diseases, reliable biofluid biomarkers capable of measuring TDP-43 pathology in living patients have been elusive.<br />
<br />
Conventional plasma TDP-43 measurements are limited by substantial biological background, as TDP-43 is expressed throughout the body and detected from numerous peripheral cell types. NeuroDex's proprietary ExoSORT&#x2122; platform addresses this challenge by selectively enriching neuron-derived extracellular vesicles from blood, providing a minimally invasive window into molecular processes occurring within neurons of the brain and spinal cord.<br />
<br />
Across multiple independent cohorts, ExoSORT-based analysis demonstrated that NDE-associated TDP-43 distinguished sporadic ALS from healthy controls. These findings were reproduced across three cohorts: 75 sporadic ALS versus healthy controls (44% increase, P=0.0003); 25 sporadic ALS versus 30 healthy controls (52% increase, P=0.002); and a third cohort of 52 ALS versus 37 healthy controls, in which NDE-associated TDP-43 was markedly elevated in disease (more than twofold higher; P<0.0001).<br />
<br />
Longitudinal analyses further showed that NDE-associated TDP-43 increases over time as disease progresses. In the most recent cohort, NDE-associated TDP-43 rose over time in sporadic ALS and tracked clinical status. Within-patient longitudinal modeling estimated an increase of approximately 45% per year in sporadic ALS, and TDP-43 levels were associated with disease severity as measured by the ALSFRS-R. <br />
<br />
"TDP-43 is emerging as a central pathology in neurodegeneration, yet measuring it in biofluids has been a long-standing challenge. A blood-based readout of TDP-43 could enable better patient selection and treatment monitoring and make clinical trials more efficient. These data are encouraging and support moving into clinical validation," said Dr. Ajay Verma, CEO of Twilight, which works with NeuroDex to develop TDP43 biomarkers for ALS and FTD. <br />

TDP43 can also serve as a treatment response biomarker. In a recent phase 2b randomized, placebo- controlled clinical trial (PARADIGM) conducted by NeuroSense Therapeutics (Nasdaq: NRSN) with its lead product candidate PrimeC (Cudkowicz et al. 2026), NeuroDex applied its ExoSORT workflow to serial samples collected longitudinally from study participants across all scheduled study visits. NDE-associated TDP-43 was measured in serial blood samples obtained throughout the trial, enabling assessment of biomarker trajectories over time in both placebo-treated and actively treated participants.<br />
<br />
In this trial, NDE-associated TDP-43 increased over time in the placebo group, consistent with the natural-history pattern observed in NeuroDex's prior studies. In contrast, participants receiving PrimeC demonstrated a statistically significant reduction in NDE-associated TDP-43 from baseline over the 6-month treatment period.<br />

Notably, the biomarker analyses were derived from the same randomized clinical dataset that demonstrated favorable effects on functional decline, survival, and additional disease-relevant biomarkers. Taken together with NeuroDex's prior work, these findings strengthen the evidence linking NDE-associated TDP-43 to ALS disease biology and support its continued evaluation in clinical studies as a pharmacodynamic biomarker.<br />

First runner up is Polk Springs Morgan owned and bred by Grant and Sally Rogers. Second runner up is Channel Acres Cantbelieveitsnotbutr owned and bred by George, Susan and Hannah Neal.<br />

Nationally, purchase loan closing costs declined by 2.9%, driven largely by falling home prices, which reduced transfer tax burdens across many markets. In total, 28 states saw closing costs decrease, while 23 states experienced increases.<br />

The most dramatic single-market shift occurred in D.C., where closing costs dropped 21.1%. Because D.C. has one of the highest transfer tax rates in the country, the significant decrease in the average purchase price here had a compounding effect on closing costs, though the area still has the highest dollar-value closing costs nationally. Conversely, home prices in Delaware rose modestly, pushing closing costs up by 4.5%, keeping it the most expensive state as a percentage of sale price at 3.06%.<br />

Other key findings from LodeStar's 2024 v. 2025: Year-Over-Year Mortgage Closing Cost Report include:<br />

* An 7.8% surge in refinance volume, with refi closing costs averaging less than half of purchase closing costs;<br />
<br />
* Higher-than-average refinance closing costs for New York and Florida borrowers, driven by taxes structured around the loan or note amount, rather than a property transfer, which then applies to both purchase and refinance transactions; and<br />
<br />
* A growing trend of recording fees being redirected to fund non-real-estate programs, such as affordable housing and homelessness services, with little to no borrower visibility.<br />

"The connection between closing costs and housing affordability is often overshadowed by other components to the equation, like interest rates and down payments," said Ron Carvalho, director of data operations at LodeStar. LodeStar's dataset is drawn from the company's closing cost calculator platform, which serves mortgage lenders across the United States. Each record represents a distinct quote (not a funded loan), identified by a unique search ID. Where lenders run multiple quotes for the same loan, only distinct transactions are counted.<br />

Closing costs are reported both inclusive and exclusive of recording fees and transfer taxes, as these components are highly jurisdiction-specific and can create misleading cross-state comparisons when included without context. The report presents averages by state; individual market results may vary based on loan amount, property value, transaction type and title provider selection.<br />

ASC Direct installed its first AOPACK BM2508-Plus in 2020. Following several years of production experience with the machine, the company ordered an additional BM2508-Pro and BM1800-Mini in 2025. The two new machines were commissioned in March 2026 through GTS Europe, AOPACK's sales and service partner in the United Kingdom.<br />

The expanded installation allows ASC Direct to assign different orders to the machine best suited to the required board size, box format and production quantity.<br />

FROM ONE AOPACK MACHINE TO THREE<br />
<br />
ASC Direct produces bespoke corrugated packaging for both customers ordering directly through its online system and customers purchasing corrugated cartons through traditional offline sales channels. Its online ordering system allows customers to configure custom-sized cardboard boxes in quantities ranging from 25 to 2,000 units. According to GTS Europe, more than 90 percent of these online orders are for quantities of 100 boxes or fewer.<br />

This production profile requires frequent order changes, flexible box dimensions, efficient board utilization and careful control of labor and setup costs.<br />
<br />
ASC Direct had already gained practical experience with its BM2508-Plus before investing in two additional AOPACK models.<br />
<br />
The BM2508-Pro and BM1800-Mini were both added primarily to increase ASC Direct's overall production capacity. The three AOPACK machines can produce a broad range of corrugated box styles, allowing the customer to distribute orders according to available capacity, board width, required functions, and production schedules.<br />

In addition to increased capacity, the BM2508-Pro provides functions including cold gluing, QR code scanning and glue flap crushing, helping ASC Direct integrate more production steps into one coordinated workflow.<br />

The BM1800-Mini also includes cold gluing and QR code scanning. It processes narrower corrugated sheets than the BM2508-Pro, but its compact footprint allows the customer to add production capacity while using less factory floor space.<br />

FLEXIBLE CORRUGATED BOX PRODUCTION<br />

AOPACK box making machines combine key corrugated converting processes within a programmable production workflow.<br />
<br />
Depending on the machine model, configuration and required box style, these processes can include slitting, creasing, slotting, horizontal cutting, customized shape cutting, gluing and printing.<br />
<br />
Operators can enter the required board dimensions and box specifications through the machine control interface, allowing the equipment to adjust for different production orders without requiring a traditional cutting die for each box size.<br />

This approach is particularly useful for short-run production, where frequent size changes and small quantities can make conventional tooling and multi-stage production less economical.<br />

The AOPACK box making production workflow helped ASC Direct reduce inefficiencies associated with its previous rotary slitter process for short-run orders. The machines can slit, score and cut corrugated board in one coordinated process while supporting a wide range of packaging formats.<br />

The decision follows a multi-year contraction across the wine industry. The downturn has hit growers hardest, who bear the fixed costs of farming regardless of demand.<br />
<br />
"We can't take a year off the vineyard. We've got to prune, water, and take care of it, and a huge amount of that is manual labor," explained Bewley. "At some point, the numbers have to pencil. Right now, they don't - not because of the vineyard, but because the market for this kind of fruit isn't there yet."<br />
<br />
First planted in 1993, Alder Springs, whose fruit has supplied many of California's most acclaimed and adventurous wineries, became known for doing what few believed possible. Bewley pioneered organic frost protection to grow high-quality fruit on a rugged mountain site at elevations between 1,700 and 2,700 feet; committed to no-till farming from the outset to protect the salmon-bearing streams that run through the property; and spent 15 years transitioning the vineyard to regenerative practices. Today, it grows 43 commercial varieties - among them rarities in California such as Petite Arvine, Mencía, Counoise, Pelaverga and Aligoté - alongside ongoing trials of hundreds of variety, clone, and rootstock combinations.<br />
<br />
Over the years, Alder Springs fruit appeared in a wide range of highly rated wines from dozens of significant California producers.<br />
<br />
Looking ahead, Bewley will redirect his focus to other profitable enterprises, including his carbon-sequestration company, Carbon Storage Solutions LLC, land-stewardship work, and his role on the board of the Pacific Forest Trust. Some members of the Alder Springs team will transition to those businesses alongside him; the vineyard is assisting others in finding new opportunities in the industry.<br />

Bewley will retain ownership of the ranch with no intention to sell. <br />
